Lipidomic Characterization of Body Fluids and Tissues for Patients Suffering from Serious Diseases Using Mass Spectrometry
Postdoc researcher will participate in the development and introduction of new methods in the lipidomic characterization of body fluids and tissues for patients suffering from serious human diseases, such as cardiovascular diseases, various types of cancer or Alzheimer disease. The lipidomic characterization includes all analytical steps starting from the preparation of biological samples (plasma, surgically removed cancer tissues and other types of samples), the preparative fractionation of individual lipid classes and finally detailed characterization using one or multidimensional liquid chromatography coupled to mass spectrometry. The lipidomic quantitation will be performed for large number of lipids followed by the multivariate statistical evaluation (PCA, PLS, etc.) with the goal to find biomarkers for individual types of studied diseases.
Mass Spectrometry Imaging of Biological Tissues
Postdoc researcher will participate in the development of new methods for mass spectrometry imaging using matrix-assisted laser desorption/ionization (MALDI) and other related desorption ionization techniques (e.g., LDI, NALDI, DESI). Individual analytical steps will be carefully optimized, mainly the sample preparation, the way of matrix introduction (spraying vs. sublimation procedures), the type of matrix and finally the parameters of mass spectrometric measurements. Mass spectrometry imaging will be applied especially for the analysis of drugs and their metabolites in animal tissues and for the spatial imaging of lipid distribution in patient and animal tissues.
